Technical Information
Oligos are delivered lyophilized in individual vials or in 96 or 384 well plates. Each oligo is accompanied by a spec sheet listing your sequence name, sequence, MW, G/C content, Tm, OD, mg and nanomole yield. At no extra charge, every oligo is deprotected and desalted to remove small molecule impurities and is quantitated twice by UV spectrophotometry to provide an accurate measure of yield. Mass yield (OD) varies with length and decreases for shorter oligos. Any synthesis not meeting the yield guarantee is remade. In addition, we statistically sample 100 nmole oligos and perform Gel Electrophoresis. Routinely, every oligo synthesized at the 250 nmole scale or larger and all modified oligos 100 nmole or larger (standard and/or PAGE purified) are individually analyzed by Gel Electrophoresis. MALDI-TOF Mass Spec is run on all modified, purified and plate oligos up to 50 bases in length. IDT can provide you with this documentation for $25 per oligo. Analytical RP-HPLC and/or Capillary Electrophoresis of your oligonucleotide is also available for $25/sample. Additional Purification is recommended for cloning, mutagenesis, assays requiring high sensitivity and all oligos greater than 40 bases in length. Please see our web site or catalog for recommendations on additional purification. We also have an extensive selection of modifications available including dual labeled fluorescent probes for real time PCR, Custom RNA and Antisense Oligos. Our technical support staff of experienced scientists if available for free consultation about your DNA application at (800) 328-2661.

Technical Information
Millipore Sigma oligos are synthesized using our patented AbacusTM platform which yields a higher coupling efficiency than most competitor platforms. We offer 4 levels of purification: desalt, reverse phase cartridge (RP1), PAGE and reverse phase HPLC. Each and every oligo is physically tested by gel electrophoresis and spectrophotometer analysis. Additionally, each synthesis run is statistically tested using MALDI-ToF mass spectrometry analysis. At Sigma-Genosys, quality is not just a goal – it is a guarantee. In addition to our stringent quality analysis, you will receive a Quality Assurance Certificate that includes quantitated yield, melting temperature, MW and µg/OD with every oligo. You will also receive an additional label that can be used in your records or on an additional vial. All this at no extra charge.

Technical Information
LTI Custom Primers are synthesized on a highly automated computer controlled parallel array synthesizer using standard phosphoramidite chemistry. Each order is double entered into the system ensuring correct input. Coupling efficiencies are monitored throughout synthesis on each oligonucleotide by trityl analysis. Five levels of purity are currently available: deprotected only; deprotected/desalted by C18 spin-column chromatography; cartridge; HPLC and PAGE. A comprehensive certificate of analysis accompanies each oligonucleotide indicating quantity, molecular weight, extinction coefficient, sequence, and melting temperature.
